Adenosine Diphosphate Ribose Pyrophosphohydrolase in Rabbit Tissues

Abstract
|

|
In previous works from this laboratory we have described thei properties of adenosine diphosphate-5¢¥ -ribose (ADP-ribose) pyrophosphohydrolace from rabbit erythrocytes, which catalyzes the hydrolysis of ADP-ribose to yield AMP and ribose-5¢¥ -phosphate.
In the present investigation it was found that the enzyme is widely distributed in rabbit tissues other than erythrocytes, such as the kidney, liver, spleen, heart, pancreas, lung, brain, adrenal gland, and even in the serum. Accordingly, atte mpts were made to characterize the tissue enzymes for their Mgz+ requirement, Km values for ADP-ribose, optimal pH, heat stability, and their behavior toward adenine nucleotides.Evidence is presented indicating that the kidney and serum enzymes which are Mg".-independent are distinct from the Mgz+ dedependent liver and erythrocyte enzymes in their properties.It seems likely that the enzyme is one of the tissue-specific isozymes.
